How is Pfast different from a full Pfenex program?

Like the name, Pfast is fast. Our advanced Pfenex strain engineering program is historically used when a protein expressed in other platforms has been challenging for a multitude of reasons and developability of the protein is at risk. A full Pfenex platform screen takes 10 - 15 weeks, but Pfast accelerates this process by focusing on high-potential toolbox subsets, delivering faster results more efficiently.

Up to 1,000 unique expression strains generated by combining an array of hosts with a set of expression vectors, exploring a number of genetic elements with your gene of interest.
A pooled ligation strategy where your gene of interest is combined with up to 200 different vector combinations, and transformed into one host.
Screening
Duplicate samples of each clone in an automated 96 well format.
Serially diluted cell pool grown in an automated 96 well format.

Program Deliverables:

  • Detailed protein titer and analysis of soluble and insoluble fractions via SDS-CGE or ELISA provided in PowerPoint format.
  • Analytical assessments via Western blot and/or BLI for additional costs. Optional advanced analytics, including HPLC, LC-MS, LC-MS/MS, MALDI-TOF (peptide mapping) and others are performed in-house.
  • Sample material for further testing provided if suitable titers are achieved.
  • Strategic optimization recommendations of identified clone(s) for additional strain improvements, or advancement to 2L scale fermentation.
